A transcriptomic approach of salinity response in the euryhaline teleost, Dicentrarchus labrax ArchiMer
Boutet, Isabelle; Ky, Chin Long; Bonhomme, Francois.
Euryhaline teleosts possess the capacity to osmoregulate under various environmental conditions (freshwater to hypersaline water). This physiological capacity is generally monitored using enzyme activity assays (Na+/K+-ATPase...), hormones quantification (prolactine, growth hormone) or their mRNAs expression. To date, few studies addressed the genetic correlates of adaptation to varying salinity at a molecular level in such fish. In the sea bass Dicentrarchus labrax, genetic differentiation was observed at specific allozyme loci between lagoon- and open-sea populations. In the present study, we investigated transcriptomic response of D. labrax to salt- and freshwater acclimation in two organs involved in osmoregulation, gill and intestine. By using...

Aquaculture en système clos : estimation des coûts de production pour l'élevage du bar et du turbot ArchiMer
Lavenant, M; Paquotte, Philippe.
Two major constraints to aquaculture have raised in most countries : the little availability of convenient sites because of the competition for the use of the coastline and the progressive implementation of regulations to reduce the impact of the farms on the environment. The technique of recirculation, widely used for hatcheries, has been developed to answer these constraints and to get free of the natural variations in order to regulate the résults. The ongrowing experimental facilities have proved good enough results to make it possible to conceive industrial projects for sea-bass and turbot. In first approach, the production costs of these systems are similar to traditional techniques but expected productivity gains due to an increase of zootechnical...

Calcium response of KCl-excited populations of ventricular myocytes from the European sea bass (Dicentrarchus labrax): a promising approach to integrate cell-to-cell heterogeneity in studying the cellular basis of fish cardiac performance ArchiMer
Ollivier, Helene; Marchant, James; Le Bayon, Nicolas; Servili, Arianna; Claireaux, Guy.
Climate change challenges the capacity of fishes to thrive in their habitat. However, through phenotypic diversity, they demonstrate remarkable resilience to deteriorating conditions. In fish populations, inter-individual variation in a number of fitness-determining physiological traits, including cardiac performance, is classically observed. Information about the cellular bases of inter-individual variability in cardiac performance is scarce including the possible contribution of excitation-contraction (EC) coupling. This study aimed at providing insight into EC coupling-related Ca2+ response and thermal plasticity in the European sea bass (Dicentrarchus labrax). A cell population approach was used to lay the methodological basis for identifying the...

Cardiac preload and venous return in swimming sea bass (Dicentrarchus labrax L.) ArchiMer
Sandblom, Erik; Farrell, Anthony; Altimiras, Jordi; Axelsson, Michael; Claireaux, Guy.
Cardiac preload (central venous; pressure, Pcv), mean circulatory filling pressure (MCFP), dorsal aortic blood pressure (P-DA) and relative cardiac output (Q) were measured in sea bass (Dicentrarchus labrax) at rest and while swimming at 1 and 2 BL s(-1). MCFP, an index of venous capacitance and the upstream venous pressure driving the return of venous blood to the heart, was measured as the plateau in Pcv during ventral aortic occlusion. Compared with resting values, swimming at 1 and 2BL s(-1) increased Q (by 15 +/- 1.5 and 38 +/- 6.5%, respectively), Pcv (from 0.11 +/- 0.01 kPa to 0.12 +/- 0.01 and 0.16 +/- 0.02 kPa, respectively), MCFP (from 0.27 +/- 0.02 kPa to 0.31 +/- 0.02 and 0.40 +/- 0.04 kPa, respectively) and the calculated pressure gradient for...

Characteristics of fads2 gene expression and putative promoter in European sea bass (Dicentrarchus labrax): Comparison with salmonid species and analysis of CpG methylation ArchiMer
Geay, Florian; Zambonino, Jose-luis; Reinhardt, Richard; Kuhl, Heiner; Santigosa, Ester; Cahu, Chantal; Mazurais, David.
Marine fish species exhibit low capacity to biosynthesise highly unsaturated fatty acid (HUFA) in comparison to strict freshwater and anadromous species. It is admitted that the Delta(6) desaturase (FADS2) is a key enzyme in the HUFA biosynthetic pathway. We investigated by quantitative PCR the relative amounts of FADS2 mRNA in European sea bass (Dicentrarchus labrax) in comparison with a salmonid species, the rainbow trout (Oncorhynchus mykiss L.). The analysis of the expression data was performed regarding the difference of the characteristics of a critical fragment of the fads2 gene promoter between sea bass and Atlantic salmon. The lower level of fads2 gene expression observed in sea bass suggested that fads2 gene putative promoter, which exhibited an...

Cloning and retinal expression of melatonin receptors in the European sea bass, Dicentrarchus labrax ArchiMer
Sauzet, S; Besseau, L; Perez, P; Coves, Denis; Chatain, Beatrice; Peyric, E; Boeuf, G; Munoz Cueto, J; Falcon, J.
Melatonin contributes to synchronizing behaviors and physiological functions to daily and seasonal rhythm in fish. However, no coherent vision emerges because the effects vary with the species, sex, moment of the year or sexual cycle. And, scarce information is available concerning the melatonin age, receptors, which is crucial to our understanding of the role melatonin plays. We report here the full length cloning of three different melatonin receptor subtypes in the sea bass Dicentrarchus labrax, belonging, respectively, to the MT1, MT2 and Mellc subtypes. MT1, the most abundantly expressed, was detected in the central nervous system, retina, and gills. MT2 was detected in the pituitary gland, blood cells and, to a lesser extend, in the optic tectum,...

Comparative effects of long-term hypoxia on growth, feeding and oxygen consumption in juvenile turbot and European sea bass ArchiMer
Pichavant, Karine; Person, Jeannine; Le Bayon, Nicolas; Severe, Armelle; Le Roux, Annick; Boeuf, Gilles.
When juvenile turbot Scophthulmus maximus and sea bass Dicentrarchus labrax were fed to satiation, growth and food intake were depressed under hypoxia (3.2 +/- 0.3 and 4.5 +/- 0.2 mg O-2 l(-1)). However, no significant difference in growth was observed between fishes maintained in hypoxia and fed to satiation and fishes reared in normoxia (7.4 +/- 0.3 mg O-2 l(-1)) and fed restricted rations (same food intake of fishes at 3.2 mg O-2 l(-1)). Routine oxygen consumption of fishes fed to satiation was higher in normoxia than in hypoxia due to the decrease in food intake in the latter. Of the physiological parameters measured, no significant changes were observed in the two species maintained in hypoxia. This study confirms the significant interaction between...

Contribution of dietary arginine to nitrogen utilisation and excretion in juvenile sea bass (Dicentrarchus labrax) fed diets differing in protein source ArchiMer
Tulli, F; Vachot, Christianne; Tibaldi, E; Fournier, Vincent; Kaushik, Sadasivam.
The role of dietary arginine in affecting nitrogen utilisation and excretion was studied in juvenile European sea bass (Dicentrarchus labrax) fed for 72 days with diets differing in protein sources (plant protein-based (PM) and fish-meal-based (FM)). Fish growth performance and nitrogen utilisation revealed that dietary Arg surplus was beneficial only in PM diets. Dietary Arg level significantly affected postprandial plasma urea concentrations. Hepatic arginase activity increased (P < 0.05) in response to dietary Arg surplus in fish fed plant protein diets; conversely ornithine transcarbamylase activity was very low and inversely related to arginine intake. No hepatic carbamoyl phosphate synthetase III activity was detected. Dietary arginine levels did...

Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer
Fauvel, Christian; Suquet, Marc; Dreanno, Catherine; Zonno, Vincenzo; Menu, Bruno.
A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Dietary TAG source and level affect performance and lipase expression in larval sea bass (Dicentrarchus labrax) ArchiMer
Morais, Sophia; Cahu, Chantal; Zambonino-infante, Jose-luis; Robin, Jean; Ronnestad, I; Dinis, M; Conceicao, L.
The influence of dietary TAG source (fish oil, triolein, and coconut oil) and level (7.5 and 15% of the diet) on growth, lipase activity, and mRNA level was studied in sea bass larvae, from mouth opening until day 24 and from day 37 to 52. Fish oil and triolein induced better growth in both experiments, this being significant at a higher dietary level. Coconut oil significantly decreased growth at the higher level, possibly as the result of an excessive supply of medium-chain TAG. Growth was not related to lipase specific activity, suggesting a production in excess to dietary needs. Body lipid content was positively related to dietary lipid level and was affected by lipid quality. In addition, larval FA composition generally reflected that of the diet. The...

Different survival of three populations of European sea bass (Dicentrarchus labrax) following challenge with two variants of nervous necrosis virus (NNV) ArchiMer
Barsøe, Sofie; Allal, Francois; Vergnet, Alain; Vandeputte, Marc; Olesen, Niels Jørgen; Schmidt, Jacob Günther; Larsen, Cathrine Agnete; Cuenca, Argelia; Vendramin, Niccolò.
Viral Nervous Necrosis (VNN, also called viral encephalo- and retinopathy (VER)), is a widespread disease of marine aquaculture caused by betanodavirus (or nervous necrosis virus - NNV), a segmented positive sense RNA virus, member of the nodaviridae family. VNN affects predominantly marine fish and cause significant losses to the Mediterranean fish farming industry, including the production of European sea bass (Dicentrarchus labrax). Of the four circulating genotypes of betanodavirus, red-spotted grouper NNV (RGNNV) and the reassortant genotype red-spotted grouper/striped jack NNV (RG/SJNNV) are most prevalent in the Mediterranean. Inheritable resistance against VNN has been detected in sea bass, and selective breeding could be a mean to limit this...

Effect of chronic exposure to ammonia on growth, food utilisation and metabolism of the European sea bass (Dicentrarchus labrax) ArchiMer
Dosdat, Antoine; Person, Jeannine; Coves, Denis; Dutto, Gilbert; Gasset, Eric; Le Roux, Annick; Lemarie, Gilles.
The chronic effects of exposing sea bass (average initial weight 100 g) to ammonia in water at 22 degreesC were first evaluated over a 61-day period (period 1, P1) during which nine different groups were submitted to nine ambient ammonia levels ranging from 0.014 to 0.493 mg 1(-1) NH3-N (0.53-16.11 mg 1(-1) total ammonia nitrogen (TA-N)) and fed using self-feeders. At the end of P1, the fish were starved for 10 days (P2). Their recovery capacity was tested over 43 days (P3) after which the exogenous ammonia supply was stopped in all treatments and the fish were allowed to feed. After 20 days of exposure a highly significant effect of ammonia was evident from the decrease in feeding activity, voluntary feed intake (VFI) and specific growth rate (SGR), and...

Effects of oleic acid on the high threshold barium current in seabass Dicentrarchus labrax ventricular myocytes ArchiMer
Chatelier, Aurelien; Imbert, Nathalie; Zambonino, Jose-luis; Mckenzie, David; Bois, P.
The present study employed a patch clamp technique in isolated seabass ventricular myocytes to investigate the hypothesis that oleic acid (OA), a mono-unsaturated fatty acid, can exert direct effects upon whole-cell barium currents. Acute application of free OA caused a dose-dependent depression of the whole-cell barium current that was evoked by a voltage step to 0 mV from a holding potential of -80 mV. The derived 50% inhibitory concentration (IC50) was 12.49 +/- 0.27 mu mol l(-1). At a concentration of 30 mu mol l(-1), OA significantly reduced the current density to about 45% of control values, but did not modify either the shape of the current-density voltage relationship or the apparent reversal potential. In addition, OA did not modify the voltage...

During 84 days, the effects of density on juvenile sea bass (Dicentrarchus labrax) (76 +/- 16 g) were studied in an experimental tank-based flow through system. Performance, stress response and resistance to virus infection were analysed under five stabilized rearing densities: 10, 20, 40, 70 and 100 kg m(-3). Water quality parameters (CO2, total ammonia nitrogen and O-2) were measured and maintained close to the recommended values for farmed sea bass by adjusting water renewal exchange. No significant differences were observed between density treatments, neither on stress response (cortisol) nor susceptibility to nodavirus. With regards to biological performances, the daily feed intake and specific growth rate were significantly lower in fish reared at...

Effects of temperature, stocking density and farming conditions on fin damage in European sea bass (Dicentrarchus labrax) ArchiMer
Person, Jeannine; Le Bayon, Nicolas.
This paper presents a non invasive, rapid and reliable way to quantitatively assess fin erosion in sea bass (Dicentrarchus labrax). The method is based on a visual assessment of fin profile and area loss of all fins except the anterior dorsal, on a scale from 0 to 4 in comparison with a perfect fin. The effects of stocking density (SD) and temperature on fin damage were investigated under experimental conditions (100-250 g fish). Over a 4-month period, mean erosion index (mean erosion level of all fins) was 10 times higher at 120 than at 20 kg m(-3), where most fins were undamaged. Damage was also dependent on time and oxygen concentration (35% lower at 53% O-2 saturation than at 105%). Fin condition was also affected by temperature: mean erosion index was...

Fish growth and health aspects of sea bass (Dicentrarchus labrax) reared in standard vs. high rate algal pond recirculation systems ArchiMer
Mladineo, Ivona; Bocina, Ivana; Przybyla, Cyrille; Fievet, Julie; Blancheton, Jean-paul.
Recirculation aquaculture system (RAS) is the type of land-based aquaculture production that interferes the least with the environment, particularly due to low effluent volume and overall water requirement. Water reuse, sometimes limited by the accumulation of waste products excreted by fish, can be improved by incorporating another biotechnical component in the form of high rate algal pond (RAS+HRAP), thus adding to the economy and self sustainability of the system. Our goal was to evaluate whether rearing fish for 1 year in a RAS or RAS+HRAP led to differences in fish production and health, by assessing: (1) fish growth; (2) impact on the morphological structure of sea bass target tissues (liver, kidney, and spleen); (3) susceptibility to monogenean...

Inter- and intraspecific crossbreeding experiments were conducted to evaluate the aquaculture potential of hybrids in the genus Dicentrarchus, focusing on fertilisation and hatching success. The experimental design consisted of 24 controlled crosses in which individual and pooled fertilisations were made between wild D. labrax individuals (8 dams and 5 sires originating from West Mediterranean and Atlantic populations) and wild D. punctatus (6 sires). Three experiments were successively performed: (1) dams from the Mediterranean population, individually considered to have good egg quality, (2) dams from Mediterranean population, individually considered to have poor egg quality and (3) crossbreeding using pooled eggs from the Atlantic population. "Freezebass" Setting up of a sperm cryobank for sea bass. Q5CT-2002-71209. Final report (01 January 2003 – 30 June 2005) ArchiMer
Chatain, Beatrice; Poline, Olivier.
Objectives : Presently, most of marine fish farmers around the Mediterranean basin grow genetically wild fish strains, which have not been genetically improved for culture performance (growth, appearance, disease resistance etc.). It is inevitable that the present gap for genetically improved strains of marine fish, and their reliable supply, will be filled. It will ultimately dominate the competitive component of marine culture industries once they evolve into commodity-style agribusiness (with fish prices being set at the lowest possible cost of production and the best quality). Those countries, which can competitively develop and supply the improved strains, will enjoy the benefit (perhaps survival) for their local industries. They will avoid...

Gene expression patterns during the larval development of European sea bass (Dicentrarchus labrax) by microarray analysis ArchiMer
Darias, Maria J.; Zambonino, Jose-luis; Hugot, K; Cahu, Chantal; Mazurais, David.
During the larval period, marine teleosts undergo very fast growth and dramatic changes in morphology, metabolism, and behavior to accomplish their metamorphosis into juvenile fish. Regulation of gene expression is widely thought to be a key mechanism underlying the management of the biological processes required for harmonious development over this phase of life. To provide an overall analysis of gene expression in the whole body during sea bass larval development, we monitored the expression of 6,626 distinct genes at 10 different points in time between 7 and 43 days post-hatching (dph) by using heterologous hybridization of a rainbow trout cDNA microarray. The differentially expressed genes (n = 485) could be grouped into two categories: genes that were...
